The IBI PCR Clean up and gel extraction kits are designed to recover or concentrate DNA fragments (50bp – 10kb) from agarose gel, PCR, or other enzymatic reaction. This method uses a chaotropic salt to dissolve the agarose gel and denature the enzymes. The DNA fragments in the chaotropic salt are bound to the glass fiber matrix of the spin column. The contaminants are washed with a wash buffer (containing ethanol) and the purified DNA fragments are eluted by a low salt elution buffer or pure water. Salts, enzymes or unincorporated nucleotides can be effectively removed from the reaction mixture without phenol extraction or alcohol precipitation. Typically, recoveries are 90 – 95% for PCR clean-up. The entire procedure can be completed in 20 minutes, and the eluted DNA is ready for use in restriction digestion, ligation, PCR, fluorescent or radioactive sequencing, and DNA labeling.
This kit will perform both PCR clean up and gel extraction procedures, eliminating the need for two kits. IBI offers these kits in 4-prep sample kits, 100 and 300 prep kits.
Replacement DF columns and collection tubes are available for use with left over reagents. Replacement DF buffer or Wash buffer is also available to replace spilled or misplaced reagents.
