Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2a format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody is specific for human CD40, a costimulatory protein on the surface of antigen presenting cells. This antibody has been shown to be cross-reactive with canine CD40.
Application Notes:This antibody was used for immunohistochemistry (IHC) of post-mortem brain tissues. In all tissues examined, the blood vessels were stained positively for CD40 (Togo et al., 2000; PMID: 11121537). This antibody was used for flow cytometry (FC) of endothelial cell lines (Lidington et al., 1999; PMID: 10638837), dendritic cells (Urban et al., 1999; PMID: 10403251), and peripheral blood B cells and monocytes (Fan et al., 2012; PMID: 22459705). This antibody was found to be the optimal antibody for CD40 marking in an FC assay of the antibodies tested (LOB7/6, 82105, 11E9, and a rabbit polyclonal antibody). Furthermore, the effect of its binding to CD40 on keratinocyte proliferation and apoptotic rate was investigated; it was found not to inhibit keratinocyte proliferation. Rather, it had the opposite effect, as it enabled the keratinocytes to retain their proliferative potential and did not induce apoptosis (Villarroel Dorrego et al., 2006; PMID: 16487107). This antibody's cross-reactivity with canine CD40 was evaluated, and it was found to be cross-reactive with canine CD40 and to be suitable for FC and immunoprecipitation (IP) (Wang et al., 2007; PMID: 17695590). As shown via SPR and FACS, this antibody's binding to CD40 overlapped with the following: antibodies 3.1.1, 21.2.1, 22.1.1, 23.5.1, 23.29.1 (classified as one group) and 23.25.1, 23.28.1, 24.2.1 (classified as another group) and the commercially available CD40L (US7288251B2).
