A 57-year-old female presented with post-transfusion purpura 5 days following a blood transfusion. Serological studies with the sera of this patient identified the presence of anti-HLA-A2. A 50-ml EDTA anti-coagulated blood sample was obtained from the patient in the acute phase of her PTP, and mononuclear cells were isolated from the blood sample by layering onto a Ficoll gradient. An oligo dT primer was used to direct first strand cDNA synthesis and IgG encoded VH was amplified and an scFv library was constructed. Phage particles expressing anti-HLA A2/A28 scFvs were selected by panning.

Chimeric Use Statement:This reformatted human antibody was made using the variable domain sequences of the original Human scFv format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:Amino acid residues 142-145 (TTKH) of the alpha2 domain of the heavy chain of the HLA class I molecules are most likely to be involved in the formation of the epitope recognised by the scfv.
Application Notes:ScFv 3PF12 has been used in immunofluorescence and flow cytometry experiments to test antibody binding to HLA-A2 positive lymphocytes and platelets (Watkins 2000). The scFv has been tested in in vitro assays and have been shown to inhibit target cell killing (Watkins 2003).
