Mice of the HCo7 HuMab Mouse(R) strain were immunized with purified recombinant IRTA5 fusion protein. Mouse splenocytes, isolated from the HuMab mice, were fused with PEG to a mouse myeloma cell line. The resulting hybridomas were then screened for the production of antigen-specific antibodies. The 2G5 clone was selected from this.

| Application Notes: | SPR was used to determine the Kd of 2G5 binding to IRTA5 as 0.028 x 10^-9 M. Using flow cytometry 2G5 was shown to be specific for IRTA5; it does not bind IRTA1, IRTA2, IRTA3, or IRTA4. Flow cytometry also showed that 2G5 directed against human IRTA-5 binds to CD 19+ B cells and the B-cell tumor lines Karpas 1106P, SU-DHL-4, Granta 519, and L-540. 2G5 does not bind CD3+ peripheral blood T cells, CD 1A+ peripheral blood dendritic cells, CD14+ peripheral blood monocytes, or CD56+ peripheral blood NK cells. |
