| Chimeric Use Statement: | This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. |
| Specificity Statement: | This antibody binds gpUL75 (gH) of the human cytomegalovirus, which is an 86-kDa glycoprotein. The heterodimer glycoprotein H-glycoprotein L is required for the fusion of viral and plasma membranes leading to virus entry into the host cell. It is though to bind a conformational epitope on the gH, and its bind is destroyed if the protein is denatured. |
| Application Notes: | This antibody successfully precipitates glycoprotein H from both HCMV infected cell lysates and HCMV-gH-VAC-infected cell lysates. Cellular localization of HCMV-gH studied by immunofluorescence on recombinant vaccinia virus and HCMV-infected cells using HCMV-16. HCMV-16 strongly neutralizes HCMV infectivity in vitro (PMID: 2831402). The reshaped humanized antibody recognized cells infected with a variety of virus strains and neutralized clinical isolates of CMV as efficiently as laboratory strains in a conventional plaque reduction assay (PMID: 9207350). |
