| Chimeric Use Statement: | This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. |
| Specificity Statement: | This antibody recognizes and binds a shared epitope of glycoprotein B (gB) of HSV1 and HSV2. |
| Application Notes: | This antibody protected mice against encephalitis and death following ocular inoculation with the virus. Fd79 (anti-gB) prevented encephalitis even when immunization was delayed until 48 hours post-infection. It significantly reduced the severity of epithelial keratitis and lowered the frequency of persistent viral infection in an outbred mouse model. Murine Fd79 has been shown to neutralize HSV-1 in vitro without complement. Both humanized and murine Fd79 cause a 90% reduction of plaques at an antibody concentration of 7 nM (1 ug/ml) (PMID: 3010559, 3030642, 2838428). The binding of antibody to HSV-infected Vero cells was tested using fluorocytometry. Immunostaining was used to view Vero cell monolayers infected with HSV-1 and total cell lysis in the presence of Fd79. Humanized version of this antibody saw a two fold decrease in affinity (PMID: 1849279). |
