Specificity Statement:This antibody is specific for BKBeta4. It shows no cross-reactivity against BKBeta1, BKBeta2 or BKBeta3. BKBeta4 is a regulatory subunit of the calcium activated potassium KCNMA1 (maxiK) channel. It modulates the calcium sensitivity and gating kinetics of KCNMA1, thereby contributing to KCNMA1 channel diversity. It decreases the gating kinetics and calcium sensitivity of the KCNMA1 channel, but with fast deactivation kinetics. May decrease KCNMA1 channel openings at low calcium concentrations but increases channel openings at high calcium concentrations. Makes KCNMA1 channel resistant to 100 nM charybdotoxin (CTX) toxin concentrations.
Application Notes:This antibody was used for immunoblotting on brain tissue from rats exposed to CVS and mouse hippocampi (Yu et al, 2018; PMID:30271963). Immunofluorescence was preformed on HEK293 cells and N2a neurons with this antibody (Chen et al, 2013; PMID:23504458). Transfected HEK293 cells were immunoblotted with this antibody (Chen et al, 2013; PMID:23504458). The hippocampi of rats were immunostained with this antibody. This assay was preformed in order to assess the integrity of mossy fiber pathway and whether MaxiK channel are expressed at mossy fiber axon and terminals (Cox et al, 2014; PMID:24486049). DRG (L4/L5) from rats was used for a western blot with this antibody (Zhang et al, 2012; PMID:22607137).
