Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2a format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody recognizes and binds phosphorylated epitopes of the Tau protein (PHF-Tau). This antibody does not cross react with normal Tau or bovine Tau. It is capable of recognizing phosphorylations at both serine 202 and threonine 205 and to some extent also serine 208 (PMID: 7624036, 26800003).
Application Notes:This antibody recognizes all isoforms from Alzheimer PHFs, but none from normal bovine or human brain. This antibody also recognizes Alzheimer's disease-associated PHF-Tau in an ELISA with no cross reactivity to normal Tau protein (PMID: 1384266). This antibody shares an overlapping epitope with another anti-tau antibody TAU1 (PMID: 1563356). This antibody strongly reacts with PHF tau in a western blot assay (PMID: 1384266, 1563356, 1376918, 1376245, 7875300). This antibody was also used for the immunohistochemical staining of different tissues of the fetal and adult human central nervous system and an Alzheimer disease patient (PMID: 8506352). This antibody was also used in an immunohistochemical autopsy study to examine p-MAPT neuropathology in the fornix and basal forebrain nuclei of 39 recently archived brain autopsies (PMID: 27793193). It was also used in the immunofluorescent detection of hyperphosphorylated tau (AT8) on a hippocampal section of an Alzheimer disease brain (PMID: 20448485).
