| Application Notes: | The scFv version of this antibody binds human B7-H1 with an affinity of 2.88 nM. The ability of this antibody to block binding of PD-1-Fc protein or B7.1-Fc protein to B7-H1-Fc was evaluated using ELISA. This antibody could effectively block B7-H1/PD-1 binding and B7-H1/B7.1 binding with an IC50 value of 1.37µg/ml and 0.521.37µg/ml respectively. The ability of the AB1 to bind to human B7-H1, mouse B7-H1 was determined by flow cytometric analysis. This antibody did not exhibit any binding to 293F cells transfected with mouse B7-H1, human PD-1 or human ICOS. This antibody was also shown to IFN-γ release in a PBMC asssay. It was also reported that this antibody can inhibit A375 human melanoma tumor growth in mouse model (WO2015195163). |
