| Specificity Statement: | The antibody is specific for human IGF-IR. It does not prevent clone 12BI from binding to the same protein. IGF-IR is a receptor tyrosine kinase which mediates actions of insulin-like growth factor 1 (IGF1). Binds IGF1 with high affinity and IGF2 and insulin (INS) with a lower affinity. The activated IGF1R is involved in cell growth and survival control. |
| Application Notes: | Specificity of this antibody was confirmed by ELISA (WO2009017679). Further, western blot was preformed to test whether the antibody cross reacts with IR protein (WO2009017679). It was also observed that the human version of this antibody was useful for antibody dependent cell cytotoxicity assays; MCF-7 or A549 target cells were incubated with this antibody and human natural killer cells. It was shown that the antibody does increase lysis of these cells (Goetsch et al, 2005; pmid:15386423). Furthermore, it was shown that the human version of this antibody is able to inhibit growth of MDA-MB-231, an oestrogen-independent breast cancer cell line in mice (Pandini et al, 2007; pmid:17451939). The human version of this antibody was also used in immunofluorescence on MCF-7 cells (Broussas et al, 2009; pmid:19165858). Finally, human version of this antibody was utilized in western blot of MCF-7 cells tumors (Broussas et al, 2009; pmid:19165858). |
