Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody is specific for NS1 protein form influenza A. NS1 inhibits post-transcriptional processing of cellular pre-mRNA, by binding and inhibiting two cellular proteins that are required for the 3'-end processing of cellular pre-mRNAs: the 30 kDa cleavage and polyadenylation specificity factor/CPSF4 and the poly(A)-binding protein 2/PABPN1.
Application Notes:To research Akt phosphorylation, western blotting with this antibody was preformed on MDCK and Vero cells infected with A/Puerto Rico/8/34, B/Lee/40 or B/Maryland/59 (Ehrhardt et al, 2007; pmid:17715214). This antibody was used to asses the viral protein expression in A549, HEK293 and Vero cells via Western blot (Kathum et al, 2016; pmid:26687707). To elucidate the ability of NS1 to bind RNA, immunoprecipitation was preformed on cell lysates from A549 cells transfected with wt NS1 or mutated versions of NS1 (Kathum et al, 2016; pmid:26687707). For analysis of NS1-RIG-I complex formation, HEK293 cells were transfected with a pCAGGS-Flag-RIG-I construct. The cell lysate was then co-immunprecipitated using this antibody (Kathum et al, 2016; pmid:26687707). Further, the co-immunoprecipitated NS1 was detected by Western blot using this antibody (Kathum et al, 2016; pmid:26687707).
