Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2b format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds the carboxy terminus of the α2-helix of class I MHC molecule and a turn on one of the underlying β-strands.
Application Notes:The binding specificity of BB7.2 towards HLA-A2 was assessed using complement-mediated cytotoxicity and cellular radio binding assay. This antibody is cytotoxic for peripheral blood leukocytes and lymphoid cell lines. The original IgG2b version of this antibody binds HLA-A2 positive cells with a binding or avidity constant of about 5 x 108 liter/mol (PMID:7035415). This antibody was used to detect HLA-A*0201-PR1 complexes using western blot and ELISA (PMID: 17493346). This antibody was also used to detect HLA-A2 complexed with pol, gag, and EBV peptides using ELISA (PMID:8562505). This antibody was used to screen human leucocyte antigen (HLA)-A2 haplotype-positive HBV patients using flow cytometry (PMID: 16108766). This antibody was also used for the immunofluorescent staining of HLA-A2.1 molecules on the cell surface of human processing defective cell line (T2) (PMID:7684681). This antibody was used in a study involving the expression of HLA class molecules in head and neck squamous cell carcinoma (HNSCC) specimens by immunohistochemistry (PMID: 9328140).
