Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format for improved compatibility with existing reagents assays and techniques.
Specificity Statement:This antibody binds an epitope comprising amino acid residues 523–527 on the E2 glycoprotein of hepatitis C virus. The hepatitis C virus is the cause of hepatitis C and some cancers such as liver cancer (hepatocellular carcinoma, abbreviated HCC) and lymphomas in humans.
Application Notes:The binding affinity of this antibody towards native and denatured recombinant E1E2 lysate was assessed using ELISA. The binding characterization of this antibody to glycoprotein E2 of HCV was analyzed by flow cytometry using the scFv version of this antibody. Further to assess the ability of this antibody to neutralize HCV a FFU assay was performed against the J6/JFH1 recombinant virus using the human IgG1 version of this antibody. The human IgG1 version was found to neutralized HCVcc of genotypes 1a, 4a and 6a however 212.1.1 could not neutralize cell-free J6 HCVcc. Finally alanine scanning was used to mutate the E2 glycoproteins. These mutated proteins were then used to map the epitope of the human version of this antibody (Keck et al, 2019; pmid:31100098).
