Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2a format for improved compatibility with existing reagents assays and techniques.
Specificity Statement:This antibody is specific for glycoprotein E2 antigenic region 412-423 of hepatitis C. The hepatitis C virus is the cause of hepatitis C and some cancers such as liver cancer (hepatocellular carcinoma, abbreviated HCC) and lymphomas in humans.
Application Notes:The binding characterization of this antibody for antigenic site 412 to 423 on the E2 glycoprotein was done using ELISA and the mouse IgG2a version of this antibody. This antibody showed strong binding for E1E2 complex from strains H77, J6E3, UKN3A1.28, UKN4.11.1, UKN5.15.7, UKN6.5.34. Further to assess the ability of this antibody to neutralize hepatitis C virus a neutralization assay on 293T cells infected with HCVpp was performed. It showed up to 90%, 77% and 60% neutralization of H77, J6E3, UKN1b12.6 strains respectively (Ruwona et al, 2014; pmid:24965471). Using bio-layer interferometry the dissociation constant of the mouse version of this antibody was determined using E2 core domain as antigen. Furthermore neutralization of virus particles was determined using a neutralization assay. This was done by trying to infect 293T cells with HCVpp in the presence of the mouse version of this antibody (Aleman et al, 2018; pmid:29954862).
