| Specificity Statement: | This antibody recognizes a conformation dependent epitope (domain B) on the E2 glycoprotein of hepatitis C virus. This antibody was reported to bind 2 regions of the E2 glycoprotein from amino acid residues 425 to 443 and aa 529 to 535 (PMID: 21813602). |
| Application Notes: | The binding characterization of this antibody towards E2 glycoprotein was done using immunofluorescence assay. The original IgG1 version of this antibody was also used in the immunoprecipitation of E2 glycoprotein expressed in 293T cells. For determination of the neutralization capabilities of this antibody towards genotype 1a HJ3-5 HCVcc infection an FFU reduction assay was performed. It was reported that this antibody could successfully neutralize HCVcc genotypes 1a, 1b, 2a, 2b, 3a, 4, 5, and 6 infections. However, HC-11, failed to neutralize genotype 5 HCVpp infection (PMID: 18400849). The epitope mapping of this antibody was done by treating the ability of the antibody to bind alanine replaced mutant E2 protein expressed in 293T cells and analyzed by ELISA (Keck et al., US8858947). This antibody was found to mediate neutralization by blocking virus binding to CD81 (PMID: 21813602). |
