The original IgG2 HC-1 antibody was isolated from peripheral blood B cells of an individual with chronic HCV genotype 1a infection who had a high serum antibody binding titer to E2 and high neutralizing activity (>1:10,000 titer) against genotype 1a HCVpp. The B cells were activated by Epstein-Barr virus and used to produce human hybridomas. The affinity-matured version of HC-1 (called HC-1AM) was created by constructing a library of yeast-displayed HC-1 single chain Fv (scFv) mutants and selecting the relevant high affinity binders.

Specificity Statement:This antibody recognizes a conformation dependent epitope (domain B) on the E2 glycoprotein of hepatitis C virus.
Application Notes:This antibody is an affinity matured version of original human IgG2 antibody HC-1. The selection of the affinity matured yeast displayed scFv mutants was done using flow cytometry. The measurement of the affinities of the resulting clones was done using BIAcore assay. The binding of the human IgG1 version of the antibody to E2 glycoprotein was determined using ELISA. The affinity matured IgG1 version of the antibody was used to neutralize a panel of HCV pseudo particles and infectious 2a HCV virus and it was found that the affinity matured version of HC-1 was able to neutralize viral isolates that was not neutralized by wild-type HC-1 (PMID: 22002064). This antibody was used in an investigation to check the spectrum of neutralization of JFH1-based HCVcc-expressing patient-derived Core-NS2 from genotypes 1a (strains H77, TN, and DH6), 1b (J4, DH1, and DH5), 2a (J6, JFH1, and T9), 2b (J8, DH8, and DH10), 2c (S83), and 3a (S52, DBN, and DH11). It was reported that HC-1AM could neutralize all 16 different HCVcc recombinants (PMID: 25043937).
