| Specificity Statement: | This antibody binds triple helical citrullinated form of C1 epitope (amino acids 359 – 369 : ARGLTGRPGDA) of collagen type II. The ACC1 antibody cross reacts with denatured CII as well as the denatured triple helical C1 epitope. C1 peptide contains two arginine which are post translationally converted to citrulline. ACC1 was found to recognize the second citrulline at position 365 (PMID: 19204106). This antibody is also was found to cross-react with several noncitrullinated epitopes on native CII (PMID: 28679953). |
| Application Notes: | Antibodies to citrulline-modified proteins have a high diagnostic value in rheumatoid arthritis (RA). The binding specificity of mouse IgG2a version of this antibody towards citrullinated protein was determined through ELISA. This antibody was also used in the immunohistochemical staining of cartilage in paw samples obtained of arthritis induced mice. This antibody was also reported to induce and mediate arthritis in mice (PMID: 19204106). This antibody was also reported to cross react with noncitrullinated epitopes on native CII, causing proteoglycan depletion of cartilage and severe arthritis in mice. A study demonstrated the molecular mechanism on how ACC1 trigger arthritis (PMID: 28679953). This antibody was also used in the preparation of a cocktail for induction of rheumatoid arthritis in mice (PMID: 32448385). |
