Quick Overview

The original antibody was isolated from a yeast display library constructed from peripheral B cells from a donor with asymptomatic chronic HCV genotype 2b infection. The donor's serum contained a high antibody binding titer (>1∶10,000) to E2 and similarly high neutralizing titers against genotype 2a JFH1 HCVcc and 1a H77C HCVpp. Affinity maturation of the original antibody was done using a yeast display approach to isolate the clones that have improved binding and neutralization activities against HCV isolates.

Chimeric Use Statement: This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG1 format for improved compatibility with existing reagents assays and techniques.
Specificity Statement: This antibody recognizes a conformation dependent antigenic epitope on the E2 glycoprotein. This antibody specifically binds the antigenic domain D of the virus comprising amino acids 441, 442, 446 and 616 (PMID: 22511875).
Application Notes: This antibody is an affinity matured version of the original murine parent antibody HC84.26. This antibody is capable of recognizing E1E2 proteins from six different HCV genotypes and subtypes in an ELISA. This antibody is capable of neutralizing 1a and 2a HCVcc isolates of strains H77C and JFH1 (PMID: 22511875; 27641232). The original antibody was also used in the immunoprecipitation of 1a H77C recombinant E1E2 lysate (PMID: 22511875). The original antibody demonstrated synergistic effects towards the majority of the HCVcc’s when combined individually with AR4A (PMID: 25043937).
