Specificity Statement:This antibody recognizes a linear epitope comprising residues 412 to 423 of HCV envelope glycoprotein E2. It specifically recognizes the antigenic domain E of the virus comprising amino acids 413, 418 and 420 (PMID: 29703495). This antibody is also capable of recognizing and neutralizing the virus bearing N417S mutation associated with a glycosylation shift from Asn-417 to Asn-415 that enables HCV to escape neutralization by antibodies such as HCV1 and AP33 (PMID: 25737449). The hepatitis C virus is the cause of hepatitis C and some cancers such as liver cancer (hepatocellular carcinoma, abbreviated HCC) and lymphomas in humans.
Application Notes:This broadly neutralizing antibody successfully neutralized JFH1 2a HCVcc and HCVcc isolates from genotypes 1a, 4a, and 5a. This antibody was found inhibit the binding of E2 glycoprotein to CD81 in a dose dependent manner. The binding affinity of the scFv version of this antibody was determined by using surface plasmon resonance in a BIAcore 3000. The binding specificity of the antibody to native or denatured recombinant E2 was determined by ELISA. Epitope mapping of the antibody was also done using ELISA (PMID: 23097455). HC33.1 was also reported to neutralize virus bearing the E2 N417S and N417T adaptive mutations with the glycosylation shift to Asn-415 (PMID: 25737449). This antibody was used in a western blot analysis to identify HEK293 and Sf9 derived sE2 (PMID: 30651366).
