| Application Notes: | The original IgM version of this antibody strongly reacted with breast cancer cells (MCF7) and colon cancer cells (HT29) in an ELISA. Immunofluorescence staining suggested that the antigen this antibody detected was located in the cytoplasm of MCF7 and HT29 cell lines but there was no positive staining detected on cervical cancer (HeLa) and fibroblast normal cells (3T3). The original IgM version of the antibody detected a 55 kDa protein that was present in the extract of MCF7 and HT29 cell lines in a western blot assay (Abdullah & Ali., 2006). The scFv version of this antibody (B7) also showed high reactivity towards MCF-7 breast cancer cells in an ELISA (PMID: 24301925). The binding reactivity of the scFv version of this antibody was also tested using flow cytometry and it was suggested that the antigen was expressed in periplasmic space of MCF-7 cells when tested using an annexin V-based apoptosis assay (PMID: 25022376). |
