The original antibody was generated by immunizing female white leghorn (Gallus domesticus) chickens with 100 μg purified α-enolase in an equal volume of Freund's complete adjuvant by an intramuscular injection. The total RNA was extracted from the enlarged spleens for scFv antibody library construction. This clone was isolated by panning the scFv library against alpha-enolase.
