| Specificity Statement: | The antibody binds to an epitope located on the head region of the hemagglutinin protein of influenza H3N2 virus. The epitope is a seven-residue long peptide WTGVTQN present as a loop near the receptor binding site. The antibody cross reacts with natural variants of the epitope which include: single mutants (RTGVTQN, WTGTTQN and WTGVTPN), double mutants (WTEVAQN, WTGVAQS, and WTGVAQD) and a triple mutant (WPEVAQN). Further, the antibody binds to wild type (HA) proteins and its variants (Mut1-WTEVAQN; Mut2-WTGVAQS; Mut3-WTGVAQD; Mut4-WPEVAQN; Mut5- RTGVTQN; Mut6-WTGVTPN; Mut7-WTGTTQN). |
| Application Notes: | The specificity of this antibody (scFv) was confirmed in ELISA analysis. The binding affinity of the scFv fragment towards the HA1 protein was found to be Kd= 0.1 μM using surface plasmon resonance. Microneutralization assay was performed to assess the neutralization activity of the scFv fragment against the influenza A virus strain A/X31(H3N2) on Madin-Darby Canine Kidney (MDCK) cells (IC50= 20 μg/ml). This antibody was used for flow cytometry on HEK293 cells (Vashisht et al, 2019; pmid:31352231). |
