Specificity Statement:This antibody binds the the central conserved region of respiratory syncytial virus (RSV) G protein of strains A2 and B1. It was reported that the antibody binds 'HFEVFNFVPCSICSNNPTCWAIC' peptide sequence in the conserved region of RSV G protein. Respiratory syncytial virus (RSV) is a major cause of severe lower respiratory tract infections in infants and the elderly. The surface of the virion is decorated with the fusion glycoprotein (RSV F) and the attachment glycoprotein (RSV G), which binds to CX3CR1 on human airway epithelial cells to mediate viral attachment and subsequent infection. RSV G is a major target of the humoral immune response, and antibodies that target the central conserved region of G have been shown to neutralize both subtypes of RSV and to protect against severe RSV disease in animal models.
Application Notes:The binding characterization of this antibody was done using Pepscan-based ELISA. This antibody can potently neutralize both subtyes of RSV A2 and B1 in the presence of complement. However, it was incapable of neutralizing both subtype in absence of complement. The original antibody has 50% inhibitory concentration (IC50) of 0.12 nM and 0.06 nM for strains A2 and B1, respectively. This antibody also demonstrated potent neutraliztion of RSV in well-differentiated HBECs cultured at an air-liquid interface, in absence of complement, with an IC50 of 4.7 nM. The Fab version of this antibody bound G peptide with a dissociation constant of 0.52 nM and secreted form of RSV G from strain A2 (sG) with equilibrium dissociation constants (KD) of 3.3 nM (PMID: 29509814).
