| Specificity Statement: | This antibody binds the central conserved region of the respiratory syncytial virus (RSV) G protein at amino acids between 1–173 and blocks the interaction of G with CX3CR1, thereby inhibiting viral attachment. This antibody binds G proteins of long, A2 and 8/60. Respiratory syncytial virus (RSV) is a major cause of severe lower respiratory tract infections in infants and the elderly. The surface of the virion is decorated with the fusion glycoprotein (RSV F) and the attachment glycoprotein (RSV G), which binds to CX3CR1 on human airway epithelial cells to mediate viral attachment and subsequent infection. RSV G is a major target of the humoral immune response. |
| Application Notes: | This antibody shows both anti-viral and anti-inflammatory effects and is a promising candidate for treating for active human RSV infection. The binding specificity of this antibody towards G protein of three strains of RSV namely long, A2 and 8/60 was determined using ELISA (PMID: 2420819). This antibody showed no neutralizing activity against RSV A2, Long, or 82-776 (PMID: 2459412). This antibody was used in a therapeutic treatment where it was found to mediate virus clearance and decrease leukocyte trafficking and interferon gamma (IFN-γ) production in the lungs of RSV-infected mice (PMID: 19264600). Another study reported that treatment with mAb 131-2G on day 3 after RSV infection reduces both inflammation and RSV titer in the lungs of RSV infected mice. It is proposed that this antibody alone or in combination with other anti-RSV F protein neutralizing antibodies could be effective in decreasing the virus-induced host response to infection and improve treatment outcome (PMID: 19545210). Combined treatment with antibodies 130-6D and 131-2G significantly decreased RSV-associated pulmonary inflammation compared to either antibody alone (PMID: 23300550). |
