| Specificity Statement: | The NMC-4 antibody binds the GP1b-α binding site on the A1 domain of human von Willebrand factor (also know as vWF or Factor VIII related antigen). vWF is a plasma glycoprotein (GP) that circulates in blood as a series of multimers, in association with factor VIII procoagulant protein. It is produced by endothelial cells and megakaryocytes and is involved in the process of hemostasis by engaging in adhesion of platelets to the sites of vascular injury. Mutations in this gene lead to Von Willebrand's disease which causes frequent bleeding. |
| Application Notes: | The binding characterization of this antibody was done using ELISA (Shima et al, 1985). This antibody was used in the initial characterization of the three distinct functional sites on the von Willebrand factor molecule (PMID: 2477370). This antibody can be used for the immunoprecipitation of the von Willebrand factor from a sample (PMID: 7679102). A study reported that this antibody was shown to inhibit its binding to GP Ib, completely abolished SIPA (sheer induced platelet aggregation) under high shear force, even in the presence of epinephrine. However, this antibody only partially inhibited SIPA under low shear force (PMID: 1360339). This antibody is also reported to completely inhibit vWF binding to platelet glycoprotein (GP) lb induced by either ristocetin or botrocetin at an IgG concentration of approximately 10 micrograms/mL, and also blocked binding of asialo-vWF to GP lb (PMID: 1984791). |
