Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG2a format for improved compatibility with existing reagents assays and techniques.
Specificity Statement:This antibody binds human CD7. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. CD7 is useful in the identification of precursor T cells as well as an important marker for the identification of leukemic T cells.
Application Notes:This antibody binds human T cell antigen CD7 and can be used for the detection of CD7 on cell surfaces using flow cytometry. This antibody can also be used for the study of CD7 expression on cells of T cell lineage using double and triple immunofluorescence assays (PMID: 2479683). The chimeric version of this antibody has been studied as a prophylactic regimen in renal allograft patients. The antibody was well tolerated, induced a modulation of the CD7 molecule, and did not trigger anti-mAb host antibodies (PMID: 2479683). This antibody was used in the treatment of patients with rheumatoid arthritis. During treatment T-lymphocyte numbers decreased and T-lymphocyte CD7 expression was absent in all but one patient (PMID: 1720991). This antibody shows potent immunosuppressive effects in vitro. The addition of RFT2 to cell cultures inhibited the T cell proliferation induced by tuberculin PPD and OKT3 but not by phytohemagglutinin, concanavalin A or phorbol myristic acid. RFT2 had to be present during the first 24 h of culture in order to elicit inhibition. Inhibition of proliferation was not due to down regulation of interleukin-2 receptor on the surface of T cells and suppressive effects could be transferred by mononuclear cells pre-treated with RFT2 (PMID: 1712681).
