Chimeric Use Statement: This reformatted human antibody was made using the variable domain sequences of the original Human IgG1 format for improved compatibility with existing reagents assays and techniques.
Specificity Statement: This antibody binds the VP1 protein of the BK virus (Human polyomavirus 1). Polyomaviruses are icosahedral, non-enveloped, double-stranded DNA viruses. They measure 40-45 nm in diameter and are comprised of 88% protein and 12% DNA. Infection with the human polyomavirus BK (BKV) is essentially ubiquitous, with estimates ranging between 80 and 90% of the population globally infected. Primary infection most often occurs during childhood (i.e., before age 10) and results in either a mild, non-specific, self-limited illness or no symptoms at all.
Application Notes: This clone can be used for the detection of BK virus in a sample via ELISA. This clone is derived from P165E2 and has ~3-fold greater affinity and ~8-fold more potent activity (EC90) on serotype II, ~10-fold greater affinity for serotype IV than the parental clone. The parental clone P165E2, binds all four serotypes of BKV but cannot recognize denatured VP1 in western blot. Binding of the antibody is affected with mutations F66A, I145A, K69A and E82A in the VP1 protein (US9862760).
