Chimeric Use Statement:This reformatted human antibody was made using the variable domain sequences of the original Human Fab format for improved compatibility with existing reagents assays and techniques.
Specificity Statement:This antibody binds human and mouse BCMA. It acts as a receptor for TNFSF13B/BLyS/BAFF and TNFSF13/APRIL. BCMA expression is highest on terminally differentiated B cells. BCMA is involved in mediating the survival of plasma cells for maintaining long-term humoral immunity. It also activates NF-kappa-B and JNK. The expression of BCMA has been linked to a number of cancers, autoimmune disorders, and infectious diseases.
Application Notes:The selection of specific binders having affinity for BCMA was done using ELISA and their affinity was determined using SPR analysis. The original Fab antibody bound human and mouse BCMA with a monovalent binding affinity of 45 nM and 74 nM respectively. The binding of the IgG version of this antibody to membrane bound human BCMA was determined by performing flow cytometry on BCMA-expressing H929 cells and MKN45 cells. The assessment of the competition between A-APRIL and this antibody was performed on H929 cells by quantifying the binding of A-APRIL in presence of increasing concentrations of this antibody and measured by flow cytometry. This antibody was reported to not block or increase APRIL-dependent NF-κB activation and also does not block or increase BAFF-dependent NF-κB activation in vitro. This antibody was also reported to be used in the generation of antibody-drug conjugates and anti-BCMA/anti-CD3 T cell bispecific antibodies for possible immunotherapy applications in treatment of multiple myelomas (EP2953972).
