| Specificity Statement: | This antibody binds human BCMA. It acts as a receptor for TNFSF13B/BLyS/BAFF and TNFSF13/APRIL. BCMA expression is highest on terminally differentiated B cells. BCMA is involved in mediating the survival of plasma cells for maintaining long-term humoral immunity. It also activates NF-kappa-B and JNK. The expression of BCMA has been linked to a number of cancers, autoimmune disorders, and infectious diseases. |
| Application Notes: | The selection of specific binders having affinity for BCMA was done using ELISA and their affinity was determined using SPR analysis. The original Fab antibody bound human BCMA with a monovalent binding affinity of 1.8 nM. The binding of the IgG version of this antibody to membrane bound human BCMA was determined by performing flow cytometry on BCMA-expressing H929 cells and MKN45 cells. The assessment of the competition between A-APRIL and this antibody was performed on H929 cells by quantifying the binding of A-APRIL in presence of increasing concentrations of this antibody and measured by flow cytometry. This antibody was reported to not block or increase APRIL-dependent NF-κB activation and also does not block or increase BAFF-dependent NF-κB activation in vitro. This antibody was also reported to be used in the generation of antibody-drug conjugates and anti-BCMA/anti-CD3 T cell bispecific antibodies for possible immunotherapy applications in treatment of multiple myelomas (EP2953972). |
