| Specificity Statement: | This antibody is specific for the envelope domain III (EDIII) of TBEV. It binds a conserved epitope near the lateral ridge of EDIII adjoining the EDI–EDIII hinge region of the E protein. The antibody recognises EDIII from all three TBEV subtypes: Western European (TBEVWE; e.g., Neudoerfl strain), Far Eastern (TBEVFE; e.g., Sofjin strain), and Siberian (TBEVSi; e.g., Vasilchenko strain). It also cross-reacts with EDIII from other tick-borne flaviviruses, including Langat (LGTV), louping ill (LIV), Omsk hemorrhagic fever (OHFV), Kyasanur forest disease (KFDV), and Powassan (POWV) viruses. |
| Application Notes: | The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody showed neutralizing activity against TBEV RVPs (IC50 <1 ng/ml) and authentic TBEV Hypr strain (a TBEVWE isolate; IC50=67.2 ng/ml). Immunofluorescence was performed on PS cells using this antibody. Prophylaxis and therapeutic experiments in BALB/c mice infected with TBEV Hypr showed the antibody was efficacious in prevention (even at low doses ~0.005 mg/kg) and early treatment of the infection (up to 3 days post-infection). The crystal structure of the Fab fragment in complex with the EDIII domains from all three TBEV subtypes was determined (Agudelo et al. 2021; PMID: 33831141). |
