The original antibody was generated by immunizing a barnevelder chicken with an antibody antigen cocktail comprising recombinant purified human antigens annexin XI, CENP-B, Topo, Jo-1, Ro60, Rpp38 and U1A in complete Freund's adjuvant. B cells that were harvested from the bone marrow and peripheral blood lymphocytes of immunized chickens was used to generate recombinant scFv phage display libraries. The specific clone was identified by panning the library against specific antigen.

| Specificity Statement: | This antibody binds human major centromere autoantigen B (CENP-B). It is a DNA-binding protein that is derived from transposases of the pogo DNA transposon family. It contains a helix-loop-helix DNA binding motif at the N-terminus, and a dimerization domain at the C-terminus. It interacts with centromeric heterochromatin in chromosomes and binds to a specific 17 bp subset of alphoid satellite DNA, called the CENP-B box. This protein is proposed to play an important role in the assembly of specific centromere structures in interphase nuclei and on mitotic chromosomes. It is also considered a major centromere autoantigen recognized by sera from patients with anti-centromere antibodies. |
