Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Human Fab format for improved compatibility with existing reagents assays and techniques.
Specificity Statement:The antibody recognizes Human OPGbp (residues 143-317). OPGbp acts as decoy receptor for TNFSF11/RANKL and thereby neutralizes its function in osteoclastogenesis. It inhibits the activation of osteoclasts and promotes osteoclast apoptosis in vitro.
Application Notes:The specificity of the original format of the antibody was confirmed by ELISA analysis. The binding affinity of the Fab and IgG1 fragments of the antibody to OPGbp was measured by surface plasmon resonance (Kd≈ 140 nM and 0.33-0.43 nM respectively). The interaction of human OPGbp and its receptor ODAR was measured in a fluorescence energy transfer (FRET) assay. The Fab fragment of the antibody inhibited OPGbp binding to soluble ODAR-Fc fusion with an IC50 of 550 nM. Bone Marrow Assay using the Fab fragment of the antibody gave a 50% decrease (IC50) at 57.8 nM. In RAW Cell Assay the Fab fragment of the antibody showed an IC50 of 15 μg/ml, 300 nM. In RAW Cell Assay, the 3 full length versions of the antibody “AT”405, “AT”406 and “AT”407 showed IC50 of 20.1 nM, 60.3 nM & 21.4 nM, respectively. “AT”405 or “AT”407 were tested in murine bone marrow assay, showing IC50 of 2.15 ug/ml (14.4 nM), and 1.85 ug/ml (12.5 nM), respectively. “AT”406 was tested in the bone marrow coculture assay and inhibited TRAP formation with an IC50 of 2.35 μg/ml (14.4 nM) (US20140147444).
