The original antibody was generated by immunizing a barnevelder chicken with an antibody antigen cocktail comprising recombinant purified human antigens annexin XI, CENP-B, Topo, Jo-1, Ro60, Rpp38 and U1A in complete Freund's adjuvant. B cells that were harvested from the bone marrow and peripheral blood lymphocytes of immunized chickens was used to generate recombinant scFv phage display libraries. The specific clone was identified by panning the library against specific antigen.

| Specificity Statement: | The antibody is specific for Jo-1. Histidyl-tRNA synthetase is a member of the aminoacyl-tRNA synthetase family, it catalyzes the ATP-dependent ligation of histidine to the 3'-end of its cognate tRNA, via the formation of an aminoacyl-adenylate intermediate (His-AMP). Jo-1 represents a key autoantibody target in the anti-synthetase syndrome that is marked by myositis as well as extra-muscular organ complications including interstitial lung disease (ILD). |
