Specificity Statement:This antibody binds human PD-L1.
Application Notes:The antibody could significantly inhibit the binding of PD-1 and PD-L1 at the cellular level. The humanized version of the antibody was constructed. The humanized version of the antibody detected the PD-L1 by western blot analysis. The binding affinity of the mouse and humanized version of the antibody to PD-L1 was measured by surface plasmon resonance, the affinity was respectively 8.81 nM 7.77 nM. In vivo studies demonstrated that the humanized version of the antibody could effectively inhibit tumour growth showing potential tumour therapeutic value (CN109232740). This antibody was also used in the generation of a bispecific antibody (BsAb) in a dual-variable-domain immunoglobulin (DVD-Ig) format targeting CD47 and PD-L1. The variable-domain from the PD-L1-blocking mAb #18 was introduced in tandem onto h4C1 to generate a dual variable domain bsAb that binds bivalently to both CD47 and PD-L1. This BsAb retained blocking activity for both interactions, but lacked the hemagglutinating activity of the parental m4C1 (Shi et al.; 2020; PMID: 32296041; Kaur et al.; 2020; PMID: 33244513).
