Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds human integrin alpha 5 beta 1 near the N-terminus. Epitope mapping of SG/19 revealed that Thr82 in the 1 subunit, located at the outer face of the boundary between the I-like and hybrid domains, was the key binding determinant for this antibody. Integrin alpha5 beta 1 is also known as fibronectin receptor and binds matrix macromolecules and proteinases and thereby stimulates angiogenesis. Integrin α5β1, is a member of the integrin family, is a heterodimer composed by two subunits, α5 (ITGA5/CD49e) and β1 (ITGB1/CD29), and both are necessary for complete biological functions. Application Notes:This antibody was capable of recognizing integrin alpha 5 beta 1 expressed on the surface of K562 cells via immunofluorescence staining. This antibody also precipitated molecules with weight of 110 and 140 kDa under nonreducing conditions from K562 cells and Ramos cells, and 130 kDa when reduced. These results were similar to the ones reported for integrin alpha 5 beta 1. The effect of this antibody on migration beneath stromal cells was investigated by inoculating Nalm-6 and BMS2 cells together with this antibody. It was seen that the presence of this antibody blocked the migration of Nalm-6 cells beneath stromal cells (PMID: 1400595).
