Specificity Statement:This antibody specifically binds the amino acid sequence 'EGLPEPLMLRWSKEGDGGIMS' in the α3 structural domain of the human HLA-G5 and HLA-G6. HLA-G5 and HLA-G6 are secreted forms of HLA-G that are non-classical major histocompatibility class Ib molecules involved in immune regulatory processes at the maternal-fetal interface. HLA-G expression has been reported in numerous types of cancer, always associated with more advanced stage and aggressive development of the tumor.
Application Notes:This antibody can bind human HLA-G peptide 'EGLPEPLMLRWSKEGDGGIMS' in an ELISA with a binding affinity of Ka= 1.01 X 10^(9) L/mol. This antibody was capable of detecting G5 and G6 isoforms of human HLA-G in a western blot and does not cross react with any other isoforms of HLA-G. This antibody was also used in the detection of soluble HLA-G5 and HLA-G6 using intracellular flow cytometry of K562-HLA-G5 and K562-HLA-G6 cells. This antibody can also be used for the immunohistochemical staining of HLA-G5/6 molecular expression in gastric cancer tissue using immunohistochemistry (CN114605543).
