Specificity Statement:This antibody binds mouse sclerostin and cross reacts with human sclerostin and recognizes an epitope adjacent to sclerostin's binding site for the Wnt co-receptor LRP6. Sclerostin is a secreted glycoprotein with a C-terminal cysteine knot-like (CTCK) domain which is produced in osteocytes (type of bone cell). The glycoprotein sclerostin has been identified as a negative regulator of bone growth that acts through inhibition of Wnt signaling and bone formation. Neutralizing anti-sclerostin antibodies are able to restore Wnt activity and enhance bone growth thereby presenting a new osteoanabolic therapy approach for diseases such as osteoporosis.
Application Notes:The binding reactivity of this antibody for mouse sclerostin was tested using ELISA. This antibody binds mouse and human sclerostin with a binding affinity of Kd= 46nM and Kd=260 nM respectively. This antibody efficiently neutralizes sclerostin's Wnt inhibitory activity. In vitro interaction analysis using sclerostin variants revealed that this neutralizing Fab binds to sclerostin's flexible second loop, which has been shown to harbor the LRP5/6 binding motif (PMID: 27558933).
