Chimeric Use Statement: This chimeric mouse antibody was made using the variable domain sequences of the original Human IgG4 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement: This antibody binds a conformational epitope which can also be formed or mimicked by the human IL-20 derived peptide comprising amino acids 101-PDHYTLRKISSLANSFLT-118. It also cross reacts with mouse IL-20. IL-20 is a pro-inflammatory and angiogenic cytokine mainly secreted by monocytes, granulocytes, dendritic cells and skin keratinocytes that plays crucial roles in immune responses, regulation of inflammatory responses, hemopoiesis, as well as epidermal cell and keratinocyte differentiation. IL-20 shows effects in several inflammatory diseases, such as rheumatoid arthritis, atherosclerosis, cancer, and liver fibrosis via regulating cytokines and chemokines. IL-20 acts on renal cells and contributes to inflammation, fibrosis, and apoptosis through its receptors IL-20R1, IL-20R2, and IL-22R1.
Application Notes: This antibody can bind a fusion protein of human IL-20 with Gaussia luciferase in a GLuc-sandwich ELISA. This antibody was capable of neutralizing recombinant IL-20 and IL-20 receptor mediated STAT3 activation and rhIL-20 and IL-20 receptor mediated induction of KZ136-NLuc Reporter Gene. The in vivo therapeutic effect of this antibody was studies using an ear inflammation assay in 8 weeks old C57BL/6J mice by intradermal injection of human cytokine IL-20. This antibody was successful in neutralizing the injected human IL-20 in a mouse model (US20170189524). The IgG4 version of this antibody binds human IL-20 with a very high binding affinity of Kd= 91fM (PMID: 27426947).
