| Specificity Statement: | This antibody binds human Interleukin-20 (IL-20). IL-20 is a pro-inflammatory and angiogenic cytokine mainly secreted by monocytes, granulocytes, dendritic cells and skin keratinocytes that plays crucial roles in immune responses, regulation of inflammatory responses, hemopoiesis, as well as epidermal cell and keratinocyte differentiation. IL-20 shows effects in several inflammatory diseases, such as rheumatoid arthritis, atherosclerosis, cancer, and liver fibrosis via regulating cytokines and chemokines. IL-20 acts on renal cells and contributes to inflammation, fibrosis, and apoptosis through its receptors IL-20R1, IL-20R2, and IL-22R1. |
| Application Notes: | The binding characterization of this antibody towards human IL-20 was done using ELISA. This antibody inhibits IL-20 induced proliferation of calf pulmonary artery endothelial cells (CPAEs) and human HUVEC cells in vitro (US7786274). The therapeutic efficacy of this antibody was also tested in a collagen-induced arthritis (CIA) rat model. In vivo, treatment with 7E alone or in combination with etanercept significantly reduced the severity of arthritis by decreasing the hind paw thickness and swelling, preventing cartilage damage and bone loss, and reducing the expression of IL-20, IL-1β, IL-6, RANKL, and matrix metalloproteinases (MMPs) in synovial tissue. Selectively blocking IL-20 inhibited inflammation and bone loss in rats with CIA (PMID: 20722035). |
