Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2b format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody binds p19 subunit of mouse interleukin-23 (IL-23). IL-23A associates with IL-12B to form IL-23 interleukin which is a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of pro-inflammatory cytokine IL-17. Mouse IL-23 also induces strong proliferation of memory T cells. IL-23 is secreted by activated dendritic cells.
Application Notes:This antibody can neutralize the bioactivity of IL-23 with no effect on IL-12 p70 bioactivity. This antibody can be used for the detection of mouse IL-23 p19 in an ELISA (PMID: 18802055). This antibody was used in assay to determine the effect of anti-IL23p19 antibody on increase in IL-17 production after neutralization of IL-4 in T cells cultured with IL-4 DCs or with non transduced DCs (PMID: 17195211). This antibody was reported to inhibit lactic acid-induced enhancement of IL-17 production by OT-II mouse splenocytes that had been stimulated with the TLR ligand peptidoglycan (PGN) and bacillus Calmette-Gue´rin cell wall skeleton (BCG-CWS) (PMID: 18490716).
