Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:The antibody is specific for CD44v6. The antibody recognises an epitope encoded by variant exon v6 of CD44. CD44v6 is a tumor associated antigen abundantly expressed in head and neck squamous cell carcinomas (HNSCC) and in normal squamous epithelium.
Application Notes:The binding affinity of the antibody to CD44v6 was measured by surface plasmon resonance. The antibody showed a specificity of tumor localization when administrated together with another anti-CD44v6 antibody in mice in in vivo experiments (Verel et al. 2002; PMID: 11992408). The specificity of the humanized version of the antibody was confirmed by ELISA anlaysis (Kd= 3.4 nM). The antibody had good therapeutic efficacy, biodistribution and tumor uptake in in vivo experiments in mice. Mice treated with 186Re-BIWA 4 showed a decreased in the tumor volume (US6972324B2). The radioactively labelled humanized version of the antibody was employed in a clinical trial in patients with advanced squamous cell carcinoma of the head and neck showing promising antitumor effects (Börjesson et al., 2003; PMID: 14506195).
