Chimeric Use Statement:This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:The antibody is specific for human-RANK-L. The antibody cross reacts with murine RANKL. RANKL is a tumor necrosis factor related protein that binds to, and activates, the TNF receptor related protein Receptor Activator of NF-kB (RANK). RANKL acts as the key cytokine that regulates osteoclast differentiation and activation during normal bone remodeling and during disease
Application Notes:The specificity of the original format of the antibody to human and mouse RANKL was confirmed by ELISA analysis (EC50= 0.26 nM and 0.067 nM, respectively). The antibody could block the RANKL binding to RANK (IC50 = 2.23 nM). The original format of the antibody could inhibit the RANKL-inducible luciferase activity (IC50 = 3.31 nM). The binding affinity of the original format of the antibody to RANKL was measured by surface plasmon resonance (Kd= 0.7 nM) (WO2019080909).
