| Application Notes: | The specificity of the original format of the antibody was confirmed by ELISA analysis (IC50 = 5.65 nM). The binding affinity of the original format of the antibody to human HGF was measured by surface plasmon resonance (Kd= 30.1 pM). The antibody could bind to non-reduced and reduced rhHGF by western blot analysis. The antibody could efficiently neutralize HGF binding to c-Met. The antibody inhibited HGF induced proliferation in 4MBr-5 cells. The ability of the original format of the antibody to inhibit tumor growth was tested in an U87MG xenograft model. The percent of inhibition of the antibody was measured to be 73%. The ability of the original format of the antibody to inhibit tumor growth was tested in an U118 xenograft model. The percent of inhibition of the antibody was measured to be 57% (US7659378B2). |
