| Chimeric Use Statement: | This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques. |
| Specificity Statement: | This antibody is specific for the DI domain, including residues D170, G171, Q174, 223-KADPPSCDLQS-233, 270-KCPPKY-275, and Q286 of rift valley fever glycoprotein N |
| Application Notes: | The original version of this antibody was used for an ELISA on rift valley fever Gn and Gc. Further this antibody was used for flow cytometry on HEK293T cells transiently transfected with plasmids of pEGFP-M, pEGFP-Gn, pEGFP-Gc or pEGFP-Gn mutants. This antibody was used for a blocking assay, the rift valley virus was preincubated with this antibody and the mixture was then added to Vero cells. This antibody was used for a western blot on its linear epitope. This antibody was used for surface plasmon resonance on rift valley fever Gn and Gc. A neutralization assay with this antibody was performed using flow cytometry on rift valley virus and Vero cells. The antibodies were added pre-infection. This antibody was used for an in vivo assay. Mice were infected with the rift valley virus. Then post infection they were given this antibody. This lead to all of the mice surviving, where the control group all died. Finally the structure of this antibody was determined using x-ray crystallography (Wang et al., 2019; PMID:30936489). |
