| Specificity Statement: | The antibody is specific for the envelopment polyprotein of SBV. It binds to a conformational epitope on the apical end of the head domain near the N686 glycosylation site. Its epitope has an area of 907 A2 and is centered on the third disulfide bond, with contributions from the regions around helices h8–h10 and h12–h13. |
| Application Notes: | The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody reacted with SBV by indirect immunofluorescence (Wernike et al., 2015; PMID: 25889366). The binding affinity of the original format of the antibody to the recombinant SBV head–stalk construct was measured by surface plasmon resonances (KD = 5.4 ± 0.3 nM). The original format of the antibody neutralized SBV on Vero cells in a focus reduction neutralization (FRNT) assay with ND50 = 0.15 ± 0.01 µM. The scFv format of the antibody was co-crystallized in complex with the SBV Gc head domain. The original format of the antibody could protect IFNAR−/− mice from lethal SBV infection in vivo (Hellert et al., 2019; PMID: 30787296). |
