Chimeric Use Statement:This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2b format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement:This antibody is specific for amino acids 53-63 of HMGB1.
Application Notes:The original antibody was used for an ELISA on recombinant rat HMGB1. This antibody was used for a western blot on recombinant rat HMGB1. The epitope of this antibody was determined using peptide binding. Mice were subjected to a cecal ligation and puncture, after which this antibody was administered. It showed an increased survival rate in these septic mice (US7632500B2). This antibody was used for immunofluorescence on RAW 264.7 cells, to study intracellular HMGB1. This antibody was used for an ELISPOT on RAW 264.7, C57 and HMC-12 cells (Wähämaa et al., 2007; PMID:16980509). This antibody was administered to mice subjected to ACLT, the antibody was able to protect cartilage (Aulin et al., 2020; PMID:31982563). This antibody was tested in two murine arthritis models, it showed a decrease in general disease severity (Schierbeck et al., 2011; PMID:21666956). This antibody was used in vivo on SOD1G93A transgenic mice, to test the effect of this antibody on ALS, the antibody did not increase survival time but it did improve early motor deficits and reduced inflammation (Lee et al., 2019; PMID:30782181).
