| Specificity Statement: | This antibody binds the severe fever and thrombocytopenic syndrome virus glycoprotein N. Severe Fever with Thrombocytopenia Syndrome (SFTS) is an emerging infectious disease caused by the SFTS virus which is transmitted to humans through the bite of an infected tick. Most patients have symptoms such as fever, low platelet count and leukopenia, and may also have symptoms such as nausea, vomiting, diarrhea, headache, muscle pain, lymphatic enlargement, and bleeding. Recently, first case of human-to-human transmission of SFTS virus was reported in Japan. |
| Application Notes: | The binding capability of this antibody for SFTSV glycoprotein N was confirmed using ELISA and SPR. The binding affinity of this antibody for SFTSV Gn was evaluated to be Kd= 3.54X10^(-10) nM. This antibody was capable of neutralizing live virus in infected Vero cells with an IC50 of 2.805μg/mL. In an in vivo animal protection experiments the INFAR -/- mice which were injected with a dose of 10 mg/kg of SF5 antibody showed a survival rate of 100%. This antibody in combination with another anti-SFTSV Gn antibody SF5 was used for the generation of a bispecific antibody. In vivo experiments with bispecific antibody increased the survival rates of animals to 100% (CN114736291; PMID:28096333). |
