| Application Notes: | The binding affinity and specificity of the original version of this antibody (mouse Fab) were determined in a competitive ELISA. Structural analysis of this antibody — free and in complex with THC — was determined through crystallization. This antibody was also used to develop a sensitive, one-step, homogeneous FRET immunoassay for detecting THC and its metabolites in saliva samples (Niemi et al., 2010; PMID: 20630472) (US8518653B2). This antibody was successfully transiently expressed in N. benthamiana by using a geminiviral expression system. The plant-produced version of this antibody retained the binding activity to Δ9-THC and its metabolites and provided high sensitivity in detecting Δ9-THC in marijuana, inflorescence, and leaf extracts but not in stem, root, or non-cannabis controls (Boonyayothin et al., 2022; PMID: 35686006). |
