Specificity Statement:This antibody binds nerve growth factor (NGF). NGF is important for the development and maintenance of the sympathetic and sensory nervous systems. NGF is a neurotrophic factor that plays a key role in neuropathic and inflammatory-induced pain, promotes hyperalgesia and allodynia.
Application Notes:This antibody showed positive reactivity with human NGF in an ELISA. The original IgG antibody was reported to bind human NGF with a binding affinity of Kd= 4pM (US7601818). Upon administration, fulranumab binds to NGF, preventing its binding to and activation of the NGF receptors TrkA and p75NTR. Inhibition of the NGF pathway may prevent the perception of pain and may induce analgesia (NCI drug dictionary). A study examined the analgesic efficacy and safety of fulranumab in adults with chronic osteoarthritis pain found that fulranumab treatment resulted in statistically significant efficacy in pain measures and physical function versus placebo (PMID: 23748114). A phase I, randomized, placebo-controlled, double-blind study reported that AMG 403 was generally safe and well-tolerated in both healthy volunteers and knee osteoarthritis patients, and exhibited linear pharmacokinetics (PMID: 26449617).
