| Application Notes: | The specificity of the original antibody (mouse IgG3) was tested via ELISA, showing strong binding activity to GD2 ganglioside. The antibody exhibited significant reactivity against neuroblastoma, brain tumor, and melanoma cell lines, as well as notable complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC) in vitro. A chimeric version of this antibody, KM1138, was created by combining the mouse variable regions with human constant regions. Compared to the original antibody, the chimeric antibody displayed equivalent binding to GD2 and similar CDC activity but demonstrated superior ADCC activity. A humanized version of this antibody, KM8138, was created and tested for its ability to retain binding specificity and cytotoxic activity. The humanized antibody maintained a strong affinity to GD2 ganglioside, exhibited comparable CDC to the original and chimeric antibodies, and surpassed its predecessors in terms of ADCC. The humanized version of the antibody was further tested in the context of a fusion protein with human IL-2 (KM8138-hIL-2), demonstrating maintained binding to GD2 and comparable hIL-2 activity, indicating that the fusion does not impair the antibody’s original properties (WO2001023573).
